SPIN - Specialized Pro-resolving lipid mediators INteractome

Specialized Pro-Resolving Mediators (SPMs) are a distinct class of bioactive lipid molecules that orchestrate the active resolution phase of inflammation, restoring tissue homeostasis without compromising host defense. Despite their therapeutic relevance, a comprehensive, structured, and machine-readable representation of SPM biosynthetic pathways, receptor interactions, biological targets, and downstream molecular signaling networks remains largely unavailable.

Over the past two decades, more than 50 structurally and functionally diverse SPMs—such as resolvins, protectins, maresins, Cysteinyl-SPMs and lipoxins—have been identified and characterized across various models of acute and chronic inflammatory diseases, including arthritis, cardiovascular disease, neurodegeneration, and metabolic disorders. To systematically capture the mechanisms of SPM action, we are developing the SPM-Interactome (SPIN) by extending biosynthetic and signaling maps from the Atlas of Inflammation Resolution (AIR) (Serhan et al., 2020. PMID: 32893032). All curated interactions are mapped to Systems Biology Graphical Notation (SBGN) to integrate them with the AIR and prepare for in silico perturbation experiment using previously designed map analysis tools (Hoch et al., 2022. PMID: 35473910). SPIN can be used to identify unique and shared biological targets across user-defined SPM subsets, making it a suitable platform for exploring SPMs in combinatorial therapies. The SPIN will provide a structured, machine-readable web resource to support systems-level analysis of inflammation resolution.
